Effect of Ethacrynic Acid on Endolymphatic DC Potential

Abstract

The effect of ethacrynic acid (Edecrin) on the endolymphatic direct current (DC) potential was studied in guinea pigs. A marked reduction in the endolymphatic DC potential was found in those animals injected with ethacrynic acid as compared with control animals. Light microscopy of the temporal bones of animals treated with ethacrynic acid showed vacuolization of the cells of the stria vascularis.

These changes were more striking in the basal turn than in the apical turns. There was marked dilation of the vessels of the stria vascularis, but there were no significant changes in the organ of Corti or the spiral ganglia on light microscopic examination. The depressive effect of ethacrynic acid on the endolymphatic DC potential may be mediated through damage to the stria vascularis.
